目的探讨 HDV 感染树嗣肝组织中 Bcl_2/Bax 表达与 HDV 感染之间的关系,以及 Bcl_2/Bax 在丁型肝炎肝细胞凋亡中的作用.方法采用免疫组化技术对45份 HDV 感染树鼩肝组织中HDAg,Bcl_2/Bax 的表达进行了检测;应用原位末端标记技术对肝细胞凋亡进行检测;并应用免疫组化双重染色对 HDAg,Bcl_2/Bax 的表达以及肝细胞凋亡进行了检测.结果 45份肝组织中有33份可检出 Bax(阳性率73%),有7份可检出 Bcl_2(阳性率16%),有41份可检出凋亡细胞(阳性率91%).HDAg 表达与 Bcl_2/Bax 表达之间有显著相关性(X_1~2=27.6,X_2~2=35.9,P<0.05),HDAg 表达越强,Bax 表达越强;相反,Bcl_2表达则越弱.凋亡在 HDAg 阳性和阴性细胞中均可发生,以在 HDAg 阳性细胞内发生为主.Bcl_2/Bax 表达与肝细胞凋亡之间有显著相关性(X_1~2=40.1,X_2~2=38.2,P<0.05),Bax 表达越强,凋亡阳性细胞越多;相反,Bcl_2表达越强,凋亡阳性细胞越少.结论丁型肝炎病毒感染和未感染的肝细胞均可发生凋亡,但凋亡只在少数细胞内发生;HDV 可诱导 Bax 表达,但对 Bcl_2表达无明显诱导作用,Bcl_2/Bax 在肝细胞凋亡中起重要作用.
Objective To investigate the relationship between Bcl_2 / Bax expression and HDV infection in HDV-infected liver tissue and the role of Bcl_2 / Bax in hepatocyte apoptosis in hepatitis D.Methods Immunohistochemistry was used to detect the expression of Bcl_2 / The expression of HDAg and Bcl 2 / Bax in liver tissue were detected. The apoptosis of hepatocytes was detected by in situ end labeling. The expressions of HDAg, Bcl 2 / Bax and hepatocyte apoptosis were detected by immunohistochemical double staining Results 33 out of 45 liver tissues were positive for Bax (positive rate was 73%), 7 were positive for Bcl_2 (positive rate was 16%), and 41 were positive for apoptosis (91% ). There was a significant correlation between Hg expression and Bcl_2 / Bax expression (X_1 ~ 2 = 27.6, X_2 ~ 2 = 35.9, P <0.05), and the stronger the HDAg expression was, the stronger the Bax expression was; Apoptosis could occur in both HDAg positive and negative cells, and mainly in HDAg positive cells.The expression of Bcl_2 / Bax was significantly correlated with hepatocellular apoptosis (X_1 ~ 2 = 40.1, X_2 ~ 2 = 38.2, P <0.05). The stronger the expression of Bax was, the more apoptotic positive cells were. On the contrary, the stronger the expression of Bcl_2 was, the less apoptotic cells were. CONCLUSIONS: Hepatitis D Both infected and uninfected hepatocytes could induce apoptosis, but apoptosis occurred only in a few cells. HDV induced the expression of Bax, but had no obvious effect on the expression of Bcl_2. Bcl_2 / Bax played an important role in hepatocyte apoptosis .
