研究利用固相萃取-高效液相色谱技术检测淡水产品中的微囊藻毒素MC-LR和MC-RR。样品经80%甲醇水溶液提取后,以固相萃取小柱进行净化,氮吹浓缩后以高效液相色谱进行分析。以65%甲醇水溶液(含0.05%三氟乙酸)为流动相,经C18反相色谱柱分离,以紫外检测器进行定量分析。线性定量范围0.1~2.0μg/g,检出限0.1μg/g。此方法操作简便、定性定量能力较强,可作为淡水产品质量检测和食品安全风险评估的可靠检测方法。
In this study, microcystins MC-LR and MC-RR in freshwater products were detected by SPE-HPLC. After the sample was extracted with 80% methanol aqueous solution, the sample was purified by solid-phase extraction cartridges and concentrated by nitrogen and analyzed by high performance liquid chromatography. A 65% aqueous methanol solution (containing 0.05% trifluoroacetic acid) as the mobile phase was separated on a C18 reversed-phase column and quantified by a UV detector. Linear range of 0.1 ~ 2.0μg / g, the detection limit of 0.1μg / g. This method is easy to operate and has high qualitative and quantitative capabilities, which can be used as a reliable test method for quality assessment of freshwater products and food safety risk assessment.
