目的:初步研究前列腺增生患者膀胱出口梗阻后逼尿肌肾上腺素能β_3受体亚型mRNA表达和作用。方法:选择前列腺增生膀胱出口无梗阻(对照组)9名和膀胱出口梗阻患者(实验组)21名,根据尿动力学将实验组分为逼尿肌稳定组和不稳定组,半定量RT-PCR方法检测三组逼尿肌β_3受体亚型mRNA的表达,离体逼尿肌拉力实验检测β_3受体激动剂对三组逼尿肌收缩力的影响。结果:对照组、逼尿肌稳定组和不稳定组β_3受体mRNA相对含量为(18.48±2.84)、(17.28±2.57)和(10.42±1.22),不稳定组β_3受体明显降低(P<0.05);β受体激动剂和β_3受体激动剂诱导逼尿肌松驰效应呈浓度依赖性,对不稳定组作用小于稳定组和对照组(P<0.05),对照组与稳定组没有差别。结论:膀胱出口梗阻逼尿肌不稳定患者β_3受体mRNA含量下降,并且对β_3激动剂的松驰反应降低,逼尿肌肾上腺素能受体β_3亚型可能参与膀胱出口梗阻后逼尿肌收缩力变化。
OBJECTIVE: To study the mRNA expression and function of adrenergicβ_3 receptor subtype in detrusor after bladder outlet obstruction in patients with benign prostatic hyperplasia. Methods: Nine patients with BPH without bladder obstruction (control group) and 21 patients with bladder outlet obstruction (experimental group) were selected. The experimental group was divided into stable group and instability group according to urodynamics. Semi-quantitative RT-PCR Methods The expression of β 3 receptor subtype mRNA in detrusor of three groups was detected. The effect of β 3 receptor agonist on contractility of detrusor in three groups was tested in vitro by detrusor tension test. Results: The relative contents of β 3 receptor mRNA in stable group and unstable group were (18.48 ± 2.84), (17.28 ± 2.57) and (10.42 ± 1.22) in control group and unstable group (P < 0.05). The effect of detrusor relaxation induced by β-agonist and β_3 receptor agonist was concentration-dependent. The effect on unstable group was less than that of stable group and control group (P <0.05), but there was no difference between stable group and control group . CONCLUSION: β3 receptor mRNA is decreased in patients with detrusor instability due to bladder outlet obstruction and the relaxation response to β3 agonist is decreased. Detrusor beta-3 subtype of adrenergic receptor may be involved in detrusor contraction after bladder outlet obstruction Force changes.
