目的探讨c-Myc基因的表达与人非小细胞肺癌细胞增殖的关系。方法体外培养H1299细胞,分为空白对照(A组)、细胞对照(B组)、脂质体(C组)、小干扰RNA对照(D组)和c-Myc小干扰RNA组(E组)。采用实时荧光定量PCR检测c-Myc mRNA表达,细胞计数试剂盒(CCK-8)法检测细胞增殖情况。结果 c-Myc siRNA瞬时转染后48h呈浓度依赖性地抑制H1299细胞中c-Myc RNA的表达。与B、C、D组比较,E组细胞增殖率降低(P<0.05)。结论低表达c-Myc基因能抑制非小细胞肺癌H1299细胞的增殖。
Objective To investigate the relationship between c-Myc gene expression and proliferation of human non-small cell lung cancer cells. Methods H1299 cells were cultured in vitro and divided into blank control (group A), cell control (group B), liposome (group C), small interfering RNA (group D) and c-Myc small interfering RNA . The expression of c-Myc mRNA was detected by real-time fluorescence quantitative PCR, and cell proliferation was detected by CCK-8 assay. Results c-Myc siRNA inhibited the expression of c-Myc RNA in H1299 cells in a concentration-dependent manner at 48h after transfection. Compared with group B, C and D, the cell proliferation rate of group E decreased (P <0.05). Conclusion Low expression of c-Myc gene can inhibit the proliferation of non-small cell lung cancer H1299 cells.
