目的 :探讨顺铂对Hca F细胞凋亡及细胞周期的影响。方法 :采用荧光 ,透射电镜及流式细胞术分析法检测肝癌细胞凋亡 ,细胞周期和增殖指数 (PI)的变化。结果 :CDDP可引起肿瘤细胞凋亡 ,并随着给药时间的延长、药物浓度的增高 ,细胞凋亡率随之增高 ,细胞凋亡形态结构越发典型 ,表现为核固缩 ,染色质边集 ,核小体形成等。同时药物浓度的升高 ,细胞周期S期比例下降 ,G0 /G1期比例上升 ,PI增殖指数下降。结论 :CDDP可引起细胞凋亡、细胞周期G0 /G1期阻滞 ,使细胞分裂增殖活动受到抑制
Objective: To investigate the effect of cisplatin on apoptosis and cell cycle of Hca F cells. Methods: Fluorescence, transmission electron microscopy and flow cytometry were used to detect the changes of apoptosis, cell cycle and proliferation index (PI) in hepatocellular carcinoma cells. Results: CDDP could induce tumor cell apoptosis. With the prolongation of drug administration, the concentration of drug increased and the rate of apoptosis increased. The morphology of apoptotic cells became more and more typical, showing nuclear pyknosis and chromatin marginalization , Nucleosome formation and so on. At the same time, the drug concentration increased, the proportion of S phase of cell cycle decreased, the proportion of G0 / G1 phase increased, and the PI proliferation index decreased. Conclusion: CDDP can induce cell apoptosis and cell cycle arrest in G0 / G1 phase, which can inhibit cell proliferation and proliferation
