目的:优选番茄皂苷A的最佳提取工艺条件。方法:采用新鲜圣女小番茄为原料,研究果胶酶用量、酶解温度、树脂量这三个单因素对番茄总皂苷得率的影响。以洗脱液量、上样流速、洗脱液浓度、吸附脂量为考察因素,应用正交试验法筛选番茄皂苷A最佳的提取工艺条件。结果:得出最佳提取工艺条件为:吸附树脂为D101大孔树脂,原料树脂量比为10∶1,上样流速4 mL/min,10倍树脂柱用量80%乙醇洗脱(体积分数),果胶酶酶解条件为:酶用量0.05%(鲜果质量分数),酶解温度为50℃,酶解时间1.5 h。结论:获得一套番茄皂苷A提取率高、简单可行的生产工艺。
Objective: To optimize the optimum extraction conditions for tomato saponin A. Methods: The fresh Lycium microphylla was used as raw material to study the effects of pectinase dosage, enzymolysis temperature and resin amount on the yield of tomato saponins. Taking the amount of eluate, the flow rate of sample, the concentration of eluate and the amount of lipids adsorbed, the optimal extraction conditions for tomato saponin A were screened by orthogonal test. Results: The optimum extraction conditions were as follows: the adsorption resin was D101 macroporous resin, the ratio of raw resin was 10: 1, the flow rate of sample was 4 mL / min, the volume of resin was 10% , Pectinase enzymatic hydrolysis conditions: enzyme dosage 0.05% (fresh fruit mass fraction), enzymatic temperature was 50 ℃, enzymatic hydrolysis time 1.5 h. Conclusion: To obtain a set of tomato saponin A high extraction rate, simple and feasible production process.
