目的 了解前列腺癌细胞雄激素受体 (AR)基因 CAG重复序列有否改变。方法 显微镜下分别取 11例前列腺癌标本的肿瘤细胞及非肿瘤细胞 ,提取 DNA,经 PCR扩增后 ,用变性胶电泳测得肿瘤细胞和非肿瘤细胞 DNA含 AR基因 CAG重复序列片段的相对长度。结果 有 3例 (2 7.3% )前列腺癌细胞 AR基因 CAG重复序列较非肿瘤细胞短。其余 8例前列腺癌细胞 AR基因 CAG重复序列与非肿瘤细胞相同。结论 一些病例的前列腺癌细胞 AR基因 CAG重复序列可发生变化 ,其变化与前列腺癌的发生、发展和对抗内分泌治疗的关系尚待进一步的研究。
Objective To investigate whether there is any change in the CAG repeat of the androgen receptor (AR) gene in prostate cancer cells. Methods Tumor cells and non-tumor cells from 11 cases of prostate cancer specimens were taken under microscope and DNA was extracted. After amplification by PCR, the relative lengths of CAG repeat fragments of AR gene in DNA of tumor cells and non-tumor cells were determined by denaturing gel electrophoresis . Results Three cases (2. 7.3%) of prostate cancer cells AR gene CAG repeat sequences shorter than non-tumor cells. The remaining 8 cases of prostate cancer AR gene CAG repeat sequence and non-tumor cells. Conclusion Some cases of prostate cancer cells AR gene CAG repeat sequence may change, its changes and the occurrence and development of prostate cancer and the relationship between endocrine therapy remains to be further studied.
