目的:探究清热败毒合剂对难愈性创面金黄色葡萄球菌、铜绿假单胞菌细菌生物膜的作用。方法:将50只SD大鼠随机分为空白对照组、清热败毒合剂组、五味清毒饮组、阿莫西林组和头孢哌酮组,每组10只,分别予生理盐水、清热败毒合剂、五味消毒饮、阿莫西林和头孢哌酮灌胃,连续3 d,制备药物血清。分光光度法检测药物血清对于金黄色葡萄球菌和铜绿假单胞菌细菌增殖的影响;结晶紫染色法和CONA-FITC/PI法检测药物血清对细菌生物膜形成的影响;生物发光法检测药物血清对于细菌自诱导分子AI-2的影响。结果:清热败毒合剂能够通过降低难愈性创面金黄色葡萄球菌和铜绿假单胞菌AI-2浓度,抑制细菌生物膜的形成,从而抑制细菌的增殖,促进创面愈合。结论:清热败毒合剂对慢性难愈性创面金黄色葡萄球菌、铜绿假单胞菌细菌生物膜的形成具有抑制作用。
Objective: To investigate the effect of Qingrexiaodu mixture on bacterial biofilms of refractory wounds such as Staphylococcus aureus and Pseudomonas aeruginosa. METHODS: Fifty SD rats were randomly divided into control group, Qingrexiaodu mixture group, Wuwei Qingdu Yin group, amoxicillin group, and cefoperazone group. Each group contained 10 rats and received saline and heat sepsis respectively. Mixtures, five flavored sips, amoxicillin, and cefoperazone were given orally for 3 consecutive days to prepare drug serum. Spectrophotometric method was used to detect the effect of drug serum on the bacterial proliferation of Staphylococcus aureus and Pseudomonas aeruginosa; crystal violet staining and CONA-FITC/PI method were used to detect the effect of drug serum on bacterial biofilm formation; bioluminescence was used to detect drug serum The effect of bacterial self-induced molecule AI-2. RESULTS: Qingrexiaodu Mixture could inhibit the formation of bacterial biofilm by reducing the concentration of AI-2 in staphylococcus aureus and Pseudomonas aeruginosa, thus inhibiting the proliferation of bacteria and promoting wound healing. Conclusion: Qingrexiaodu mixture can inhibit the formation of bacterial biofilms in Staphylococcus aureus and Pseudomonas aeruginosa in chronic refractory wounds.
