梭梭是生长在我国西北荒漠地区抗逆性极强的一种生态保护型植物。通过RT-PCR技术克隆到梭梭的转录因子HaDREB2C的cDNA全长编码区为1 125 bp(登录号为KU523927),编码374个氨基酸,其中含有一个AP2结构域;半定量RT-PCR表达量显示,该基因受干旱,高盐的诱导;将HaDREB2C克隆到酵母表达载体pGBKT7中转入AH109酵母细胞,转化子可以在SD-Trp、SD-Trp-His-Ade多营养缺陷型固体培养基中正常生长,运用β-半乳糖苷酶的滤纸分析法显色测定,证明pGBKT7下游报告基因被激活,HaDREB2C在酵母中具有转录激活活性。
Haloxylon ammodendron is an ecologically-conserved plant that grows extremely strong in the desert regions of northwestern China. The full length coding region of HaDREB2C, a transcription factor cloned into Haloxylon ammodendron by RT-PCR, was 1 125 bp (accession number: KU523927) encoding 374 amino acids with an AP2 domain. Semi-quantitative RT-PCR showed that The gene was induced by drought and high salt. HaDREB2C was cloned into yeast expression vector pGBKT7 and transformed into AH109 yeast cells. The transformants were normal in SD-Trp, SD-Trp-His-Ade multi-auxotrophic solid medium Growth, the use of β-galactosidase filter paper assay color determination, that pGBKT7 downstream reporter gene is activated, HaDREB2C in yeast transcriptional activation.
