目的:优选金银花中绿原酸微波辅助提取工艺,并对提取物的抗氧化能力进行分析。方法:以甲醇浓度、料液比、提取时间为考察因素,以提取液在327nm波长的吸光度和二苯代苦味酰基自由基(DPPH·)清除率为响应值,采用响应曲面法优选工艺;通过比较提取液对DPPH·的清除能力分析提取物的抗氧化能力。结果:最佳提取工艺为以43%的甲醇溶液作提取剂、按液料比40mL·g-1、在微波功率400W下提取8min,绿原酸提取率为7.62%;金银花提取物对DPPH·的清除能力与绿原酸含量呈正相关。结论:优化的工艺稳定、合理、可行,提取率高、提取时间短;绿原酸在金银花提取物的抗氧化能力中起主要作用。
Objective: To optimize the microwave-assisted extraction of chlorogenic acid from honeysuckle and analyze its antioxidant capacity. Methods: The concentration of methanol, the ratio of solid to liquid and the time of extraction were investigated. The absorbance at 327 nm and the scavenging rate of dibenzoyl picric acid radical (DPPH ·) were used as response values. The response surface methodology The extract was compared with DPPH · scavenging capacity analysis of the antioxidant capacity of the extract. Results: The optimum extraction process was 43% methanol solution as extractant, the extraction rate of chlorogenic acid was 7.62% at the liquid-to-liquid ratio of 40mL · g-1 and the microwave power of 400W for 8min. The scavenging capacity and chlorogenic acid content was positively correlated. Conclusion: The optimized process is stable, reasonable and feasible, the extraction rate is high and the extraction time is short. Chlorogenic acid plays a major role in the antioxidant activity of extracts of honeysuckle.
