目的 考察阿魏酸钠对氧化应激条件下 ,肝星状细胞中I型胶原及其相关调控因子CTGFmRNA表达的干预作用。方法 以 5 0 μmol·L-1H2 O2 处理的HSC -T6细胞为氧化应激模型组 ,采用RT -PCR方法检测并分析 12 5 μmol·L-1和 5 0 0 μmol·L-1SF对细胞中I型前胶原及其相关调控因子CTGF表达的影响。结果 肝星状细胞在氧化应激条件下I型前胶原mRNA和CT GFmRNA表达水平都明显上调 ,而阿魏酸钠可以下调二者的表达水平。结论 阿魏酸钠从转录水平调控胶原表达可能对肝纤维化有重要防治作用
Objective To investigate the effects of sodium ferulate on the expression of type I collagen and CTGF mRNA in hepatic stellate cells under oxidative stress. Methods HSC-T6 cells treated with 50 μmol·L-1 of O2 were selected as oxidative stress model. RT-PCR was used to detect and analyze the effects of 125 μmol·L-1 and 500 μmol·L-1 SF on cells Effect of procollagen type 1 and CTGF on its expression. Results The expression of type I procollagen mRNA and CTGF mRNA in hepatic stellate cells was significantly increased under oxidative stress, while sodium ferulate could down-regulate the expression of both. Conclusion Sodium ferulate may play an important role in prevention and treatment of hepatic fibrosis by regulating the transcription of collagen
