目的:建立用高效液相色谱法测定障眼明胶囊和片剂中葛根素和芍药苷含量的方法。方法:采用Diamonsil-C_(18)色谱柱(250 mm×4.6 mm,5μm),流动相:乙腈-0.1%磷酸溶液(13∶87),柱温20℃,流速为1.0 ml·min~(-1),检测波长为230 nm和250 nm。结果:葛根素在49.98~374.85 ng范围内线性关系良好(r=0.997 8),平均回收率为98.6%和98.7%。芍药苷在182.14~1 366.05 ng范围内线性关系良好(r=0.999 9),平均回收率为99.5%和98.7%。结论:该法简便,可靠,准确,可作为测定障眼明胶囊和片剂中葛根素和芍药苷含量的方法。
Objective: To establish a HPLC method for the determination of puerarin and paeoniflorin in Guanyingming capsule and tablet. METHODS: Diamonsil-C 18 column (250 mm × 4.6 mm, 5 μm) was used. The mobile phase consisted of acetonitrile and 0.1% phosphoric acid solution (13:87). The column temperature was 20 ℃ and the flow rate was 1.0 ml · min ~ 1), the detection wavelength is 230 nm and 250 nm. Results: The calibration curve of puerarin was linear in the range of 49.98 ~ 374.85 ng (r = 0.997 8). The average recoveries were 98.6% and 98.7% respectively. Paeoniflorin showed a good linearity (r = 0.999 9) within the range of 182.14-1366.05 ng with an average recovery of 99.5% and 98.7%. Conclusion: The method is simple, reliable and accurate and can be used as a method to determine the content of puerarin and paeoniflorin in barrier capsules and tablets.
