目的 :探讨重组人胸腺素α原快速纯化方法 ,研究它对放射损伤免疫抑制小鼠免疫功能的调节作用。方法 :经超声碎菌、离心得上清、加热分离目的蛋白 ,用苯酚和氯仿抽提 ,最后经Super dex柱层析获胸腺素α原纯品。以 4.0Gy的剂量全身一次性照射致免疫功能低下小鼠为模型 ,观察腹腔注射胸腺素α原 30 0ng/ (只·d)对免疫功能的调节作用。结果 :胸腺素α原纯品可改善损伤小鼠的脱毛、体重减轻和尾部出血斑等一般状况 ,明显提高因损伤而萎缩的胸腺重量。虽不能影响外周血白细胞的总数 ,但可提高血淋巴细胞和血小板的数目及血红蛋白的量。结论 :纯化获得的胸腺素α原具有免疫调节活性 ,可促进放射损伤后免疫功能的恢复与重建。
OBJECTIVE: To investigate the rapid purification of recombinant human thymosin α and study its regulatory effect on immune function in immunosuppressed mice with radiation injury. Methods: The supernatant was separated by ultrasonic and the supernatant was centrifuged. The target protein was isolated by heating and extracted with phenol and chloroform. Finally, the supernatant was purified by Super dex column chromatography. The mice immunocompromised at a single dose of 4.0Gy were used as the model to observe the regulatory effect of intraperitoneal injection of thymosin α at 30 ng / (d · d) on the immune function. Results: Thymosin α original product can improve the general condition of hair loss, weight loss and tail spot in the injured mice and significantly increase the weight of the thymus atrophy caused by injury. Although it can not affect the total number of peripheral white blood cells, but can increase the number of hemocytes and platelets and hemoglobin hemoglobin. Conclusion: The purified prothymosin α has immunomodulatory activity and can promote the recovery and reconstruction of immune function after radiation injury.
