目的研究体内外联合应用HSVtk/GCV系统和5-FU治疗大肠癌的疗效。方法用逆转录病毒转染人大肠癌LoVo细胞,MTT法观察GCV联合5-FU对LoVo细胞的杀伤作用。通过裸鼠皮下注射HSVtk+LoVo细胞建立人大肠癌肿瘤模型,应用GCV和(或)5-FU治疗后测量肿瘤体积计算肿瘤生长抑制率,并用HE染色观察有无器官毒副作用。结果体外实验MTT法显示HSVtk/GCV系统联合5-FU对LoVo细胞的杀伤作用较GCV组明显增强(P<0.05),体内实验表明联合治疗组肿瘤体积为(348±68)mm~3大小,明显小于其他各组(P<0.01),HE染色未观察到各器官病理改变。结论联合应用HSVtk/GCV系统和5-FU能提高HSVtk/GCV系统治疗大肠癌的疗效,且没有明显的毒副作用。
Objective To study the efficacy of combined application of HSVtk/GCV system and 5-FU in colorectal cancer in vitro and in vivo. Methods Human colorectal carcinoma LoVo cells were transfected with retrovirus. MTT assay was used to observe the killing effect of GCV combined with 5-FU on LoVo cells. Human colorectal cancer tumor models were established by subcutaneous injection of HSVtk+LoVo cells in nude mice. Tumor growth was measured after GCV and/or 5-FU treatment. Tumor growth inhibition rate was calculated, and HE staining was used to observe whether there were organ side effects. Results In vitro MTT assay showed that HSVtk/GCV system combined with 5-FU significantly increased the killing effect on LoVo cells compared with GCV group (P<0.05). In vivo experiments showed that the tumor volume of combined therapy group was (348±68) mm~3. Significantly less than other groups (P <0.01), no pathological changes in various organs were observed by HE staining. Conclusion The combination of HSVtk/GCV system and 5-FU can improve the efficacy of HSVtk/GCV system in the treatment of colorectal cancer without obvious toxic side effects.
