用差速离心和琼脂糖CL-2B柱层析方法自日本血吸虫成虫匀浆中提取和分离微粒体抗原,用ELISA法对微粒体粗抗原(JAMA-C)、尿素溶解部分(JMC)和提纯的微粒体抗原(JAMA)的敏感性和特异性进行了比较。在动力学一ELISA测定中JAMA抗原对同种抗体呈现出高的活力,达到8.8单位。试验蛔虫、肝吸虫、肺吸虫病患者血清各3份,旋毛虫病猴血清1份,结果均低于1单位,未出现交叉反应。在微量滴定板ELISA中亦呈现高敏感性和特异性,证明经过纯化,特异性活力增加。与微粒体抗原相比,胞液及尿素溶解性核抗原的活力很低。SDS-聚丙烯酰胺电泳及SDS-PAGE的电泳转移酶标免疫印斑的结果证明JAMA是由分子量14,400至200,000道尔顿以上的蛋白分子组成的不均一体,主要抗原成分的分子量在43,000道尔顿以上。
The microsomal antigens were extracted and separated from the homogenates of adult Schistosoma japonicum by differential centrifugation and agarose CL-2B column chromatography.
