目的 了解细胞周期相关调控蛋白在胰腺癌中的表达及其对胰腺癌细胞增殖的影响。方法 应用高压锅抗原修复免疫组化ABC法 ,检测 47例胰腺癌标本的细胞周期相关调控蛋白 (p5 3、p16、p2 1及Rb蛋白 )的表达 ,并用一种新的ki s5抗体检测胰腺癌细胞ki 6 7增殖指数。结果 p5 3、p16、p2 1及Rb蛋白阳性表达均位于细胞核 ,胰腺癌中 p5 3、p16、p2 1和Rb蛋白表达阳性率分别为 5 5 % (2 6 /47)、5 3 % (2 5 /47)、6 4% (35 /47)和 98% (46 /47) ;p16、p2 1及Rb蛋白表达与ki 6 7增殖指数呈负相关 ;p5 3蛋白表达与p2 1蛋白表达无关。 结论 大部分胰腺癌的异常增殖可能与 p16、p2 1蛋白阴性表达有关 ,而由Rb蛋白阴性表达导致的胰腺癌细胞的异常增殖少见。p5 3蛋白表达与 p2 1蛋白表达无关
Objective To investigate the expression of cell cycle related regulatory protein in pancreatic cancer and its effect on the proliferation of pancreatic cancer cells. Methods The expression of cell cycle related regulatory proteins (p5 3, p16, p21 and Rb proteins) in 47 pancreatic cancer specimens was detected by autoclaved antigen immunohistochemical ABC method. A new ki s5 antibody was used to detect the expression of pancreatic cancer cells Proliferation index. Results The positive expression rates of p5 3, p16, p21 and Rb protein were located in the nucleus. The positive rates of p5 3, p16, p21 and Rb in pancreatic carcinoma were 55% (2/67) and 53% (2) 5/47), 64% (35/47) and 98% (46/47) respectively. The expressions of p16, p21 and Rb were negatively correlated with the proliferation index of ki 67. The expression of p5 3 was not related to the expression of p21 . Conclusions Abnormal proliferation of most pancreatic cancers may be related to the negative expression of p16 and p21 proteins, whereas the abnormal proliferation of pancreatic cancer cells caused by the negative expression of Rb protein is rare. p5 3 protein expression has nothing to do with p2 1 protein expression
