目的探讨细胞凋亡调控基因fas、caspase-3在颞叶癫癎病人病灶内的表达及意义。方法取30例 颞叶癫癎病人手术切除的病灶,应用免疫组化的方法检测凋亡相关基因fas、caspase-3的表达,同时采用光镜、电镜 及原位末端标记(TUNEL)方法检测神经细胞凋亡。结果免疫组化染色发现fas蛋白在对照组中无表达,在癫癎 组表达明显增强(P<0.001);caspase-3在对照组中有轻微表达,在癫癎组表达亦明显增强(P<0.005)。光镜检查 及TUNEL染色均未发现凋亡的神经细胞,在进行电镜检查的10例标本中有4例发现少量早期凋亡征象的神经元。 结论癫癎病人脑内存在神经元凋亡现象。fas、caspase-3基因在这一过程中可能发挥了重要的作用。
Objective To investigate the expression of Fas and caspase-3 genes in temporal lobe epilepsy patients and their significance. Methods Thirty patients with temporal lobe epilepsy were selected for surgical resection. The expression of fas, caspase-3 was detected by immunohistochemistry. The neurons were detected by light microscopy, electron microscopy and TUNEL Apoptosis. Results The expression of fas protein was not detected in the control group, but significantly increased in the epilepsy group (P <0.001). Caspase-3 was slightly expressed in the control group and significantly increased in the epilepsy group P <0.005). Apoptotic nerve cells were not detected by light microscopy and TUNEL staining. Four of the 10 specimens examined by electron microscopy showed a small number of neurons with early apoptotic signs. Conclusion There is neuronal apoptosis in the brain of epileptic patients. fas, caspase-3 gene may play an important role in this process.
