声动力疗法刺激肿瘤微环境中CD80与CD86表达的研究

[目的]检测声动力治疗(sonodynamic therapy,SDT)对B16F10荷瘤鼠模型中肿瘤微环境T细胞共刺激分子CD80、CD86表达情况的影响。[方法]以2.0W/cm2超声强度结合250mg/kg 5-氨基酮戊酸(5-ALA)治疗B16F10荷瘤鼠,测量不同实验组小鼠体重及肿瘤大小;免疫组织化学方法检测移植瘤微环境中CD80、CD86、IFN-γ和TNF-α表达情况。[结果]与其他组相比,SDT组肿瘤体积缩小更加显著,体重变化无差异;SDT组CD80、CD86、IFN-γ和TNF-α均出现表达增高,且具有统计学意义。[结论]SDT能够加强T细胞共刺激分子的CD80、CD86的表达,促进抗肿瘤免疫的发生。 [Objective] To investigate the effect of sonodynamic therapy (SDT) on the expression of co-stimulatory molecules CD80 and CD86 on tumor microenvironment T cells in B16F10 tumor-bearing mice. [Method] The B16F10 tumor-bearing mice were treated with 2.0W / cm2 ultrasonic intensity combined with 250mg / kg 5-aminolevulinic acid (5-ALA) to measure the body weight and tumor size in different experimental groups; immunohistochemistry Environment CD80, CD86, IFN-γ and TNF-α expression. [Results] Compared with other groups, the volume of tumor in SDT group decreased more significantly and there was no difference in body weight. The expressions of CD80, CD86, IFN-γ and TNF-α in SDT group were significantly higher than those in other groups. [Conclusion] SDT can enhance the expression of T cell costimulatory molecule CD80 and CD86 and promote the anti-tumor immunity.
