动脉粥样硬化和血栓形成病理过程中,血小板与血管壁组成成分之间的交互作用起重要作用。本文研究了α-受体阻滞剂Nicergoline和β-受体阻滞剂Acebu-tolol对血小板与血管壁的弹性纤维和内皮细胞间交互作用的影响。血小板可以粘附于弹性纤维和经肾上腺素活化的组织培养的内皮细胞。作者用纯化牛轫带弹性纤维和琼酯糖混悬液填装硅化柱,经无菌盐水平衡后,再将预先与nicergoline或acebutolol温育后的PRP上柱,用0.15M盐水洗脱。用分光光度计记录洗脱的血小板峰,并作血小板计数。对照柱血小板回收率约为20~30%,(?)×10~(-4)M的nicergoline可增加回收率达40~75%,使血小板粘附于弹性纤维减少58~72%。1×10~(-6)M的
In the process of atherosclerosis and thrombosis, the interaction between platelets and components of the blood vessel wall plays an important role. This article studies the effects of the α-receptor blocker Nicergoline and β-blocker Acebu-tolol on the interaction between platelet and the elastic fibers and endothelial cells in the vascular wall. Platelets can adhere to elastic fibers and epinephrine activated tissue culture endothelial cells. The authors filled the column with purified bovine slender elastic fibers and agarose suspension and sterilized with saline. PRP preincubated with nicergoline or acebutolol was then eluted with 0.15 M saline. Eluted platelets were recorded with a spectrophotometer and platelet counts were taken. The recovery rate of platelets in the control column was about 20-30%. The recovery of nicergoline with (?) X 10 ~ (-4) M increased the recovery by 40-75%, and decreased the platelet adhesion to the elastic fibers by 58-72%. 1 × 10 ~ (-6) M
