本文对含钙硬组织制片技术及组织化学脱钙法的原理、改进方法及体会作了介绍。改进法包括:1.酸液脱钙法;2.电解脱钙法;3.离子交换树脂脱钙法;4.螯合剂脱钙法;5.组织化学脱钙法。其中改进了的电解脱钙法及离子交换树脂法,设备简易、脱钙迅速、组织损害小、染色结果满意。改进了的中性EDTA脱钙法及组织化学脱钙法,能保存细胞内显微化学成分及其活性,故用于组织化学(酶、糖原等)研究。
This article introduces the principle, improvement method and experience of calcium hard tissue preparation technique and histochemical decalcification method. Improvements include: 1. Acid decalcification; 2. Electrolytic decalcification; 3. Ion exchange resin decalcification; 4. Chelating agent decalcification; 5. Histochemical decalcification. Which improved the electrolytic decalcification and ion exchange resin method, the device is simple, rapid decalcification, tissue damage is small, the staining results are satisfactory. Improved neutral EDTA decalcification and histochemical decalcification, can save the intracellular micro-chemical composition and its activity, it is used for histochemistry (enzymes, glycogen, etc.) research.
