本文采用高效液相色谱分离并确定了去屑止痒剂1-羟基-4-甲基-6-(2,4,4-三甲基戊基)-2-吡啶酮(以下简称HMTPP)。在固定相3011-O柱上,以甲醇/水作流动相通过HPLC对保留性能进行了考察,结果表明当甲醇/水/HClO_4=75∶25∶0.05(V/V),在UV300nm检测波长下,可实现HMTPP的酸式总量测定。HMTPP的线性范围为2.0~92.0μg/ml,检测限为0.805μg/ml,酸式离解常数K_a=3.05×10-8。本方法具有分析速度快,重现性好等特点。
In this paper, high performance liquid chromatography was used to separate and identify the antidotal agent 1-hydroxy-4-methyl-6- (2,4,4-trimethylpentyl) -2-pyridone (HMTPP). Retention performance was investigated by HPLC on a stationary phase 3011-O column using methanol / water as the mobile phase. The results showed that when methanol / water / HClO 4 = 75: 25: 0.05 (V / V) Under the wavelength, the acid total amount of HMTPP can be measured. The linear range of HMTPP was 2.0 ~ 92.0μg / ml, the detection limit was 0.805μg / ml, the acid dissociation constant K_a was 3.05 × 10-8. The method has the advantages of fast analysis, good reproducibility and so on.
