为了解河南省猪瘟病毒近2年的分子特征及变异情况,本试验使RT-PCR方法从猪瘟病料中获得了5株猪瘟病毒的E2基因全序列和其中10株E0基因全序列,并对猪瘟病毒阳性样品的E2和E0基因进行测序及变异分析。结果显示:扩增的河南省5株猪瘟病毒的E2基因,其中4株猪瘟病毒E2基因与石门株E2基因相比存在14个氨基酸的变化,1株和石门株E2基因存在4个氨基酸变化。与Shimen株相比,河南省猪瘟病毒的E2基因有1个糖基化位点的突变。扩增的10株猪瘟病毒的E0基因,发现河南省的猪瘟病毒之间E0基因同源性高,且其E0基因从密码子程度上一直在向远离经典毒株Shimen株和疫苗株HCLV的方向发展。对E2基因和E0基因进行进化分析发现河南省猪瘟病毒承受净化选择压力。以E2基因为基础进行分型,发现河南省共同存在猪瘟病毒1.1型和2.1b型;提示河南省猪瘟病毒变异种类更加多样化。
In order to understand the molecular characteristics and variation of classical swine fever virus in Henan Province in recent two years, we obtained the full-length E2 gene of five CSFV strains and the complete sequence of 10 E0 genes from classical swine fever virus by RT-PCR , And the E2 and E0 genes of CSFV positive samples were sequenced and analyzed for variation. The results showed that there were 14 amino acid changes in the E2 gene of 5 classical swine fever virus strains in Henan Province and 14 E2 genes in 4 strains of classical swine fever virus and 1 gene in E2 gene of Shiimen strain and 4 amino acids Variety. Compared with the Shimen strain, the E2 gene of Henan province classical swine fever virus has a mutation in the glycosylation site. E0 gene amplification of the 10 strains of classical swine fever virus, Henan Province, found that E0 gene homology between swine fever, and its E0 gene from the codon level has been far away from the classical strain Shimen strain and vaccine strain HCLV The direction of development. The E2 gene and E0 gene evolution analysis found that Henan Province Classical swine fever virus to withstand purification pressure. Based on the E2 gene typing, it was found that the co-existence of classical swine fever virus type 1.1 and type 2.1b in Henan province suggested that the variation types of classical swine fever virus in Henan province were more diversified.
