目的:研究青蒿素对紫外线照射后小鼠表皮c-kit和PAR-2蛋白的表达,探讨青蒿素光保护作用的可能机制。方法:30只BALB/C小鼠分为3组,正常对照组,给予生理盐水灌胃;模型对照组及青蒿素组分别给予生理盐水及青蒿素灌胃后进行UVB照射。各组灌胃均为100mg/(kg.d),每日1次,持续2周。UVB照射每次剂量1 J/cm2,每日1次,共2周,累积剂量14 J/cm2,采用免疫组化检测各组小鼠表皮中干细胞因子受体(c-KIT)和蛋白酶激活受体-2(PAR-2)表达量的变化。结果:UVB反复照射后,青蒿素组皮肤中c-kit和PAR-2表达水平均明显低于模型对照组(P<0.05),且c-kit和PAR-2表达水平呈正相关性(P<0.05)。结论:青蒿素可明显抑制UVB反复照射小鼠皮肤中c-kit和PAR-2的表达,提示系统性应用青蒿素对UVB致小鼠皮肤光损伤有保护作用。
OBJECTIVE: To study the expression of c-kit and PAR-2 protein in mouse epidermis after artemisinin exposure, and to explore the possible mechanism of artemisinin photoprotection. Methods: Thirty BALB / C mice were randomly divided into three groups: normal control group and normal saline group. Rats in model control group and artemisinin group were given normal saline and artemisinin respectively and irradiated with UVB. Each group gavage were 100mg / (kg.d), once daily for 2 weeks. UVB irradiation at a dose of 1 J / cm2, once daily for 2 weeks, the cumulative dose of 14 J / cm2, immunohistochemical detection of mouse epidermis in each group of stem cell factor receptor (c-KIT) and protease activation by Body-2 (PAR-2) expression changes. Results: The expression of c-kit and PAR-2 in artemisinin group was significantly lower than that in model control group (P <0.05), and the expression of c-kit and PAR-2 was positively correlated with UVB irradiation <0.05). CONCLUSION: Artemisinin can significantly inhibit the expression of c-kit and PAR-2 in the skin of mice repeatedly irradiated with UVB, which indicates that systemic administration of artemisinin has a protective effect on skin photodamage induced by UVB in mice.
