核酸分子杂交是分子生物学研究中最重要的基本技术之一。其中,膜上印迹转印杂交则是最常用的一种分子杂交方法。它不仅在基因定位、定性和定量分析上极为常用,而且是分子克隆、基因突变及基因诊断上不可缺少的关键技术。当前,southern转印印迹技术虽然从经典的虹吸印迹法发展到电转印法、真空印迹法等,但仍有许多不能令人满意之处,例如:操作不便,需要特殊的仪器设备,每次转印结果仅供一次杂交之用等。为了克服上述不足,同时考虑到精简步骤和适应于教学,作者在实践中不断摸索、总结及参考国外经验,对常用的虹吸法southern印迹技术进行了改良。实验证明该方法简便、经济实用,适合基层应用。
Hybridization of nucleic acid molecules is one of the most important basic techniques in molecular biology. Among them, the blot blot hybridization is the most commonly used molecular hybridization method. Not only is it extremely common in gene mapping, qualitative and quantitative analysis, it is also an indispensable key technique in molecular cloning, gene mutation and gene diagnosis. At present, southern blot printing technology has developed from classical siphon blotting method to electrotransfer printing method and vacuum blotting method, but there are still many unsatisfactory places, for example, inconvenient operation, special instrument and equipment, The result is only for one hybridization. In order to overcome these shortcomings, taking into account the streamlining of steps and adapt to teaching, the author constantly explore in practice, sum up and reference to foreign experience, the commonly used siphon Southern blot technology has been improved. Experiments show that the method is simple, economical and practical, suitable for grassroots applications.
