目的:建立血浆中牡荆素浓度的分析方法,测定牡荆素血浆蛋白结合率。方法:采用平衡透析法透析,利用HPLC测定血浆中牡荆素浓度,计算牡荆素与人血浆蛋白结合率,并比较大鼠和人血浆中牡荆素的血浆蛋白结合率。结果:在2~16 mg.L-1,药物浓度对血浆蛋白结合率无显著影响,但牡荆素在大鼠和人的血浆蛋白结合率存在显著性差异,牡荆素与人的血浆蛋白结合率高于与大鼠血浆蛋白结合率。结论:牡荆素与血浆蛋白具有较强的结合。
Objective: To establish a method for the determination of vitexin in plasma and to determine the plasma protein binding rate of vitexin. Methods: Dialysis was performed by equilibrium dialysis. The plasma concentration of vitexin was measured by HPLC. The protein binding rate of vitexin and human plasma was calculated. The plasma protein binding rate of vitexin in rat and human plasma was compared. Results: There was no significant difference in plasma protein binding rate between 2 mg · L-1 and 16 mg · L-1, but the plasma protein binding rate of vitexin in rats and humans was significantly different. Vitexin and human plasma protein The binding rate is higher than that of rat plasma protein. Conclusion: Vitexin and plasma protein have strong binding.
