目的研究淋巴瘤细胞株的髓细胞白血病1(myeloid cell leukemia-1,Mcl-1)在ONO-AE-248刺激的中性粒细胞(polymorpho nuclear neutrophils,PMN)非凋亡程序化细胞死亡中的作用,为进一步确定这种新型细胞死亡模式提供线索。方法 Ficoll法新鲜分离健康志愿者外周静脉血中性粒细胞,与ONO-AE-248共同培养运用流式、RT-PCR和Western-bloting技术,对接受ONO-AE-248刺激的PMN的Mcl-1的表达进行检测。结果 ONO-AE-248刺激2 h时中性粒细胞Mcl-1 mRNA的表达明显上调,特异性抗Mcl-1单克隆抗体通过Western blotting技术检测发现其在凋亡组与非凋亡组的表达有明显差异性,表现为上升的趋势。结论 Mcl-1在ONO-AE-248诱导的非凋亡性程序化死亡中蛋白表达的上调可能是中性粒细胞凋亡和ONO-AE-248诱导的中性粒细胞非凋亡性程序化细胞死亡的关键性差异点。Mcl-1在ONO-AE-248诱导的非凋亡性程序化死亡和传统凋亡中发挥的调控作用可能是不相同的。
Objective To investigate the effect of myeloid cell leukemia-1 (Mcl-1) on the apoptosis of apoptotic cells in ONO-AE-248 stimulated polymorphonuclear neutrophils (PMN) Role, to provide clues to further determine this new model of cell death. Methods Ficoll method was used to isolate peripheral blood neutrophils from healthy volunteers and co-cultured with ONO-AE-248. Conclusion The up-regulation of Mcl-1 protein expression in non-apoptotic programmed death induced by ONO-AE-248 may be apoptosis of neutrophils and non-apoptotic programmed neutrophil programmed by ONO-AE-248 The key point of cell death. The regulatory role of Mcl-1 in non-apoptotic programmed death and traditional apoptosis induced by ONO-AE-248 may not be identical.
