目的:研究土贝母鲜品二氯甲烷提取物(DEFT)对绿色荧光蛋白标记的人三阴性乳腺癌MDA-MB-231-GFP裸鼠模型中移植瘤生长和肺转移的作用.方法:应用外科原位移植方法,建立人乳腺癌裸鼠移植模型,随机分成3组,阴性对照组(0.9% NaCl)、低剂量组(DEFT 50 mg/kg)和高剂量组(DEFT 75 mg/kg),每组5只,灌胃给药,1次/l,连续58 d.每周2次荧光成像系统采集肿瘤图像,系统自带软件计算肿瘤面积;给药结束时,称取肿瘤质量,荧光成像系统观察肺转移情况.结果:给药前后3组裸鼠体质量变化差异均无统计学意义(P>0.05).给药第14天,3组肿瘤面积比较,差异有统计学意义(P<0.05).实验结束时,土贝母低剂量组和高剂量组肺转移的发生均低于阴性对照组(P<0.05).土贝母低剂量组和高剂量的肿瘤质量低于阴性对照组,但差异无统计学意义(P>0.05)结论:鲜土贝母二氯甲烷提取物对人三阴性乳腺癌MDA-MB-231-GFP裸鼠模型中肿瘤生长及肺转移的发生有一定抑制作用.“,”Objective:To study the effect on the growth of transplanted tumor and lung metastasis in a green fluorescent protein marking human triple-negative breast cancer MDA-MB-231-GFP nude mice model when treated with the dichloromethane extract of fresh Tubeimu (Rhizoma Bolbostemmae) (DEFT).Methods:The nude mice model was established by surgical orthotopic transplantation method.The mice were randomly divided into 3 groups (5 mice in each group):negative control group (0.9% sodium chloride),low dose group (DEFT,50 mg/kg) and high dose group (DEFT,75 mg/kg).Each group was treated with oral administration once a day for 58 days.The tumor image was collected by the fluorescence imaging system twice each week,and the software was used to calculate the tumor area.At the end of the treatment,the tumor weight was recorded and the lung metastasis was observed by the fluorescence imaging system.Results:Weight changes between the control group and the treatment groups were not significantly different before and after.The tumor area between three groups began to grow significantly differently after 14 days (P<0.05).Lung metastasis rates decreased significantly at the end of the experiment (P<0.05).The treatment groups showed less tumor weight than the control group,but there was no significant difference (P>0.05).Conclusion:The dichloromethane extract of fresh Tubeimu (Rhizoma Bolbostemmae) had some inhibitory effect on the growth of malignant tunors and the occurrence of lung metastasis in human triple-negative breast cancer MDA-MB-231-GFP nude mice model.
