目的研究七叶皂苷A的大鼠血浆蛋白结合率。方法在体外用平衡透析法模拟七叶皂苷A体内与血浆蛋白结合的过程,用高效液相色谱-质谱联用(HPLC-MS/MS)法测定低、中、高3个浓度的七叶皂苷A及其转化产物七叶皂苷C在透析袋内血浆中和透析袋外缓冲液中的浓度,计算七叶皂苷A的血浆蛋白结合率。结果低、中、高3个浓度的七叶皂苷A在体外与大鼠的平均血浆蛋白结合率分别为95.8%,96.6%和95.9%,3个浓度之间的血浆蛋白结合率比较差异无统计学意义(P>0.05)。结论七叶皂苷A与大鼠血浆蛋白高度结合。
Objective To study the plasma protein binding rate of aescine A in rats. Methods The process of in vivo binding of aescine A to plasma proteins was simulated by equilibrium dialysis in vitro. The contents of aescine in low, medium and high concentrations were determined by HPLC-MS / MS. A and its conversion product aescin C in the dialysis bag plasma and dialysis buffer concentration in the buffer to calculate the aescine A plasma protein binding rate. Results The average plasma protein binding rates of aescine A at three concentrations of low, middle and high in vitro were 95.8%, 96.6% and 95.9%, respectively. There was no statistical difference in plasma protein binding rate among the three concentrations Significance (P> 0.05). Conclusion Aescin A is highly associated with plasma protein in rats.
