目的:研究肝郁脾虚证大鼠模型肝脏的差异基因表达。方法:大鼠随机分为正常对照组、肝郁脾虚证模型组2组,每组12只。采用慢性束缚+过度疲劳+饮食失节法,连续28d造模,正常对照组不予处理。于制模第29d,处杀2组大鼠,待测肝脏全基因探查。结果:肝郁脾虚证模型大鼠肝脏差异表达的基因59个,其中上调基因27个,下调基因32个。在上调的27个基因中,目前已知的功能基因有Scd1、Dbp、Upp2、Slc28a2、Sds、Hpn、Gldc、Tm7sf2、Gcat。结论:肝郁脾虚证存在异常基因表达,涉及脂类代谢、糖代谢等多个方面。提示多个基因的异常表达可能是中医肝郁脾虚证的分子基础。
Objective: To study the differential gene expression in the liver of rat model of liver-qi and spleen-deficiency syndrome. Methods: The rats were randomly divided into normal control group, model group of liver depression and spleen deficiency group 2, 12 rats in each group. Chronic restraint + fatigue + diet disorders, continuous modeling for 28 days, the normal control group did not deal with. At the 29th day of model establishment, two groups of rats were killed, and the whole liver of the liver was probed. Results: There were 59 differentially expressed genes in liver of rats with liver depression and spleen deficiency syndrome, including 27 up-regulated genes and 32 down-regulated genes. Among the up-regulated 27 genes, the currently known functional genes are Scd1, Dbp, Upp2, Slc28a2, Sds, Hpn, Gldc, Tm7sf2, Gcat. Conclusion: Abnormal gene expression exists in liver-qi stagnation and spleen deficiency syndrome, involving lipid metabolism, glucose metabolism and many other aspects. Tip abnormal expression of multiple genes may be the molecular basis of traditional Chinese medicine liver spleen deficiency syndrome.
