目的观察人工发酵虫草菌丝体干粉(CM)对糖尿病大鼠心肌核因子-κB(NF-κB)和转化生长因子-β1(TGF-β1)表达的影响。方法健康雄性SD大鼠53只,鼠龄8周,体重(220±20)g。采用数字表法将大鼠随机分为正常对照组(NC,n=10)、单纯糖尿病组(n=10)、CM小剂量组[n=12;CM 0.6g/(kg.d)灌胃]、CM中剂量组[n=11;CM 2.5g/(kg.d)灌胃]、CM大剂量组[n=10;CM 5g/(kg.d)灌胃]。单次尾静脉注射STZ(45mg/kg)造模,成模8周后处死大鼠。采用免疫组织化学法、RT-PCR法检测心肌NF-κB、TGF-β1蛋白和mRNA的表达水平。结果单纯糖尿病组心肌NF-κB、TGF-β1蛋白和mRNA表达均上调(P<0.05),CM干预可下调NF-κB、TGF-β1蛋白和mRNA的异常表达(P<0.05)。结论心肌组织中,NF-κB和TGF-β1表达的改变可能参与了糖尿病心肌病(DC)的发生,CM可能通过调节NF-κB和TGF-β1在心肌的表达发挥对心肌的保护作用。
Objective To observe the effects of Cordyceps sinensis mycelium dry powder (CM) on the expression of nuclear factor-κB and TGF-β1 in diabetic rats. Methods 53 healthy male Sprague Dawley rats were aged 8 weeks and weighed 220 ± 20 g. The rats were randomly divided into normal control group (NC, n = 10), simple diabetic group (n = 10), CM low dose group [n = 12; CM 0.6g / (kg · d) ], CM middle dose group [n = 11; CM 2.5g / (kg.d)), high dose CM group [n = 10; CM 5g / (kg.d)). A single tail vein injection STZ (45mg / kg) modeling, 8 weeks after the model was sacrificed rats. The expression of NF-κB, TGF-β1 protein and mRNA in myocardium was detected by immunohistochemistry and RT-PCR. Results The expression of NF-κB and TGF-β1 protein and mRNA in myocardium of diabetic rats were both increased (P <0.05). The expression of NF-κB and TGF-β1 protein and mRNA were down-regulated in CM group (P <0.05). Conclusions The changes of NF-κB and TGF-β1 may play a role in the development of diabetic cardiomyopathy (DC) in myocardium. CM may play a protective role on myocardium by regulating the expression of NF-κB and TGF-β1 in myocardium.
