目的探讨筋脉通对糖尿病大鼠坐骨神经组织自噬相关蛋白Beclin1和LC3的影响。方法 STZ诱导建立糖尿病大鼠模型,随机分为正常对照(NC)组、糖尿病模型(DM)组和筋脉通治疗(JMT)组。灌胃治疗16周后,Western blot法检测Beclin1、LC3蛋白表达,实时荧光定量PCR法检测Beclin1-mRNA和LC3-mRNA的转录。结果与NC组比较,DM组大鼠坐骨神经Beclin1、LC3蛋白表达和Beclin1-mRNA、LC3-mRNA的转录水平均明显降低(P<0.01);JMT组大鼠坐骨神经Beclin1、LC3蛋白和Beclin1-mRNA的转录均较DM组升高(P<0.05,P<0.01)。结论筋脉通可纠正糖尿病大鼠坐骨神经组织的自噬异常。
Objective To investigate the effect of Jinmaitong on autophagy-related proteins Beclin1 and LC3 in sciatic nerve of diabetic rats. Methods STZ induced diabetic rat models were randomly divided into normal control (NC) group, diabetic model group (DM) and JMT group. The expression of Beclin1 and LC3 protein was detected by Western blot and the mRNA of Beclin1-mRNA and LC3-mRNA were detected by real-time fluorescence quantitative PCR after 16 weeks of intragastric administration. Results Compared with NC group, Beclin1 and LC3 protein expression and Beclin1-mRNA and LC3-mRNA transcription of sciatic nerve were significantly decreased in DM group (P <0.01). Beclin1, LC3 protein and Beclin1-mRNA Transcription were higher than DM group (P <0.05, P <0.01). Conclusion: Jinmaitong can correct autophagy in diabetic sciatic nerve tissue.
