目的研究人类性别决定基因(sexdeterminingregionontheYchromosome,SRY)表达蛋白对下游基因的调节作用。方法将SRY基因HMG结构域片段与表达载体PET-15b-进行重组,构建了SRY基因的表达质粒PETSY,转入大肠杆菌中表达,组氨酸结合树脂柱纯化表达蛋白,与苗勒氏管抑制因子基因(Mülerianinhibitingsubstance,MIS)启动子区片段进行凝胶电泳阻滞试验及竞争试验。结果获得SRY基因表达蛋白,分子量接近20.8U(21kD),凝胶电泳阻滞试验及竞争试验表明,SRY表达蛋白能特异结合位于19号染色体的MIS基因的启动子区。结论提示SRY基因对MIS基因转录具有调节作用。
Objective To study the regulation of downstream genes by the expression of sexdetermining regioseontheychromosome (SRY). Methods The SRY gene fragment was cloned into PET-15b- vector. The expression vector of PETY gene was constructed and transfected into Escherichia coli. Histidine-binding resin column was used to purify the expressed protein. (Mülerianinhibitingsubstance, MIS) promoter fragment was subjected to gel electrophoresis retardation test and competition test. Results The SRY gene was expressed at a molecular weight close to 20.8U (21kD). The gel electrophoresis block test and competition test showed that SRY protein could specifically bind to the promoter region of MIS gene located on chromosome 19. Conclusion suggests that SRY gene has a regulatory effect on MIS gene transcription.
