利用正交试验法优化碳源、硝酸盐、磷酸盐、铁盐、微量元素5个因子对代谢糖脂类生物表面活性剂菌种(SLT01)产量的影响,确定了各因子的最佳配比为:碳源:石蜡8.0ml;硝酸盐:NaNO31.0g;磷酸盐:KH2PO48.0g、K2HPO42.4g;MgSO42.0g。以及微量元素MnSO40.015g和FeSO40.02g;酵母膏1.0g;H2O1000mL,得出最适合该菌种生长代谢的培养基。并通过苯酚——硫酸法对发酵产物进行定量检测,确定发酵液中糖脂类生物表面活性剂的含量。
The effects of carbon source, nitrate, phosphate, iron salt and trace elements on the production of SLT01 were optimized by orthogonal test. The optimal ratio of each factor was determined As follows: Carbon source: Paraffin wax 8.0 ml; Nitrate: NaNO 31.0 g; Phosphate: KH2PO48.0 g; K2HPO42.4 g; MgSO42.0 g. And trace elements MnSO40.015g and FeSO40.02g; yeast extract 1.0g; H2O1000mL, to obtain the most suitable for the growth and metabolism of the culture medium. The content of glycolipids biosurfactant in the fermentation broth was determined by phenol-sulfuric acid method.
