目的观察连花清瘟胶囊水溶解物与头孢替唑钠联合用药对金黄色葡萄球菌生物膜的体外破坏作用。方法药物分为头孢替唑钠组、连花清瘟组、联合用药组和空白对照组;微量肉汤稀释法分别测定连花清瘟胶囊水溶解物和头孢替唑钠对金黄色葡萄球菌的最小抑菌浓度;用酶标仪检测金黄色葡萄球菌生物膜内活菌数变化;用扫描电镜观察金黄色葡萄球菌生物膜内细菌受药物作用后的形态和大小。结果连花清瘟组活菌数较空白对照组低(P<0.05);头孢替唑钠组与空白对照组相比差异无统计学意义(P>0.05);联合用药组活菌数低于单独用药组(P<0.05)。扫描电镜显示头孢替唑钠组生物膜中,大量细菌密集在一起,但密度较空白对照组稀疏,细菌形态基本正常,大小基本一致;连花清瘟组生物膜中形态不规则,大小不一致;而联合用药组形态更加不规则,大小更加不一致。结论连花清瘟胶囊水溶解物对金黄色葡萄球菌生物膜具有显著破坏作用,与头孢替唑钠联合应用可起到协同增效的作用。
Objective To observe the effect of Lianhua Qingwen Capsule combined with ceftezole sodium on the in vitro destruction of Staphylococcus aureus biofilm. Methods The drugs were divided into ceftezole sodium group, Lianhua Qingwen group, combination group and blank control group. The contents of water soluble matter of Lianhua Qingwen capsule and ceftezole sodium against Staphylococcus aureus The minimum inhibitory concentration (MIC) was determined by microplate reader. The change of viable count in Staphylococcus aureus biofilm was detected by microplate reader. The morphology and size of bacteria in S. aureus biofilms were observed by scanning electron microscopy. Results The number of viable cells in Lianhuaqing plague group was lower than that in control group (P <0.05). There was no significant difference between ceftezole sodium group and blank control group (P> 0.05) Treatment alone group (P <0.05). Scanning electron microscopy showed that a large number of bacteria were densely packed in the biofilms of ceftezole sodium group, but their density was sparse compared with the blank control group. The morphology of the bacteria was basically normal and the size was basically the same. The shape and size of the biofilms in the Lianhuaqingwen plague group were inconsistent. The combination group more irregular shape, the size is more inconsistent. Conclusions Lianhua Qingwen capsule water lysate has a significant disruptive effect on the biofilm of Staphylococcus aureus, and the combination of ceftezole sodium and synergistic effect may play a synergistic effect.
