以精子的活率、寿命和运动状态为参数,探讨了花鲈鲜精在不同盐度、酸碱度和离体条件下的生理特性,并对花鲈精液进行超低温冷冻保存研究。结果表明:盐度和酸碱度主要影响精子的寿命,对精子的活率影响不大;精子离休后置于室温(15~17℃)下8h,其活率和寿命随时间的延长而呈下降趋势,运动状态也逐渐减弱;不同组成成分的基础溶液以及抗冻保护剂的种类、浓度和使用方法均影响精液超低温冷冻保存效果;精液冷冻前经30min降温平衡(0~2℃),效果好于直接冷冻;冻精采用水浴快速解冻法(38~40℃)的效果优于室温(15.5~17.5℃)解冻法。
The physiological characteristics of fresh perch under different salinities, pH and in vitro conditions were discussed with the sperm viability, longevity and exercise status as the parameters, and the cryopreservation of perch periwinkle was studied. The results showed that salinity and pH mainly affected the life of sperm and had little effect on sperm motility. After sperm were retired, they were placed in room temperature (15 ~ 17 ℃) for 8 h, and the survival rate and life span decreased with the prolongation of time , The exercise state also gradually weakened; different composition of the basic solution as well as the type of antifreeze protective agent, concentration and method of use affect the cryopreservation of sperm cryopreservation; sperm freezing 30min before cooling (0 ~ 2 ℃), the effect is better than Direct freezing; thawing semen using water bath quick thawing method (38 ~ 40 ℃) is better than the room temperature (15.5 ~ 17.5 ℃) thawing method.
