研究选用籽粒蛋白质含量差异显著的5个品种(系),比较分析品种间GS同工型基因在灌浆过程中籽粒和叶片表达特性及启动子序列与结构特点。结果表明,不同品种间OsGS同工型基因表达量差异显著;OsGS1;1和OsGS1;2基因在籽粒和叶片中大量表达;OsGS1;3和OsGS2基因分别仅在籽粒和叶片中大量表达;在灌浆过程中籽粒和叶片中OsGS同工型基因m RNA表达量均呈单峰曲线变化,灌浆前期m RNA表达量平稳升高,中期出现峰值,后期快速降低,且高蛋白质含量品种m RNA表达量高于低蛋白质含量品种。籽粒蛋白质含量差异显著品种间OsGS同工型基因启动子序列同源性高达99%以上;品种间有性杂交子代OsGS同工型基因启动子序列可发生碱基变化,但启动子核心元件无变化。在调控元件数量和功能上OsGS同工型基因启动子保守性很强,启动子在结构和功能上不易发生遗传变异,籽粒OsGS同工型基因m RNA表达量变化受OsGS基因启动子调控影响较小。
Five varieties (lines) with significant differences in grain protein content were selected to study the expression characteristics of the GS and its isoforms and the promoter sequences and structural characteristics of the GS isozymes in the grain filling process. The results showed that there was significant difference in OsGS isoforms expression among different cultivars. OsGS1; 1 and OsGS1; 2 genes were abundantly expressed in kernels and leaves; OsGS1; 3 and OsGS2 genes were highly expressed in seeds and leaves respectively; The m RNA expression level of OsGS isoforms in grain and leaf showed a single peak curve, and the expression of m RNA increased steadily in the early grain filling stage, peaked in the middle stage and rapidly decreased in the late stage, and the m RNA expression level of the high protein content variety was high In low protein content varieties. The homology of the OsGS isoform gene promoter among the significant cultivars with significant differences in grain protein content was over 99%. The promoter of OsGS isozymic promoter in the hybrids between cultivars had a base change, Variety. OsGS isoform gene promoter is highly conserved in the number and function of regulatory elements, the genetic variation of the promoter is not easy to occur in structure and function, and the change of m RNA expression level of OsGS isoform is influenced by the regulation of OsGS promoter small.
