目的利用DNA条形码技术对泉州口岸常见麻蝇进行鉴定,以期解决麻蝇形态鉴定存在的缺陷问题,以作为形态鉴定的补充。方法通过对泉州口岸常见的5种麻蝇进行PCR扩增、序列测定,进而进行同源性对比、分析进化距离及构建进化树。结果 5种麻蝇细胞色素氧化酶辅酶Ⅰ(cytochrome oxidase subunitⅠgene,COⅠ)基因序列除短角亚麻蝇外,其余种类与Gen Bank中已知序列的同源性均在99%以上;种内差异为0.0%~0.6%(<2%),种间差异则为5.1%~10.0%;进化树显示相同种能聚集在一起,可信度达99%。结论以COⅠ基因作为分子标记可对泉州口岸5种麻蝇进行鉴定,可作为形态鉴定的重要补充。
OBJECTIVE: To identify the common flies in Quanzhou port by using DNA barcoding technology in order to solve the defects existing in the morphological identification of flies, and to supplement the morphological identification. Methods Five species of flies common to Quanzhou port were amplified by PCR and sequenced. Then homology comparison, evolutionary distance analysis and phylogenetic tree analysis were carried out. Results The sequences of the five COI genes of the flies were 99% more homologous to the known sequences in GenBank, except for the H. tabaci. The intraspecies divergence was 0.0 % ~ 0.6% (<2%), while the interspecies difference was 5.1% ~ 10.0%. The phylogenetic tree showed that the same species could be clustered together with a confidence level of 99%. Conclusion COⅠ gene can be used as a molecular marker to identify five species of flies in Quanzhou Port, which can be used as an important complement to morphological identification.
