目的 :探讨WH5 0 5 (丹参和山楂提取物 )对实验性血栓形成的影响及其作用机制。方法 :用电刺激大鼠颈总动脉和结扎大鼠下腔静脉造成血栓形成模型 ;用放免法和聚合酶链反应方法检测体外培养血管内皮细胞缺氧后ET 1的表达。结果 :WH5 0 5剂量依赖性地延长电刺激大鼠颈总动脉后血栓形成时间 ,与对照组比较有显著差异。WH5 0 5明显抑制静脉血栓的形成 ,抑制率分别为 12 .6 4%、19.30 %和 5 7.32 %。体外培养人血管内皮细胞经缺氧后 ,细胞内ET 1mRNA表达增加 ,培养液中ET 1浓度升高。在缺氧前 ,用不同浓度WH5 0 5与血管内皮细胞共孵育 ,则可明显抑制细胞内ET 1mRNA表达 ,减少内皮细胞分泌ET 1。结论 :WH5 0 5具有抗大鼠体内血栓形成的作用 ,其作用机制可能与保护血管内皮细胞损伤有关。
Objective : To investigate the effect of WH5 0 5 (Salvia miltiorrhiza Bge. and Radix Salviae Miltiorrhizae extract) on experimental thrombosis and its mechanism of action. METHODS: The thrombosis model was induced by electrical stimulation of the common carotid artery in the rat and the ligation of inferior vena cava in rats. The expression of ET 1 in vascular endothelial cells after hypoxia was detected by radioimmunoassay and polymerase chain reaction. RESULTS : WH5 050 dose-dependently prolonged the thrombus formation time after electrical stimulation of the common carotid artery in rats. There was a significant difference compared with the control group. WH5 05 significantly inhibited the formation of venous thrombosis, and the inhibition rates were 12.6%, 19.0%, and 5.7%, respectively. After culturing human vascular endothelial cells in hypoxia, the expression of ET 1 mRNA in the cells increased and the ET 1 concentration in the culture medium increased. Before hypoxia, co-incubation with different concentrations of WH5 05 and vascular endothelial cells can significantly inhibit the expression of intracellular ET 1 mRNA and reduce the secretion of ET 1 by endothelial cells. Conclusion : The effect of WH5 05 on thrombosis in rats may be related to the protection of vascular endothelial cells.
