目的 探讨辐射对皮肤表皮生长因子受体 (EGFR)表达的影响。方法 采用免疫组化链霉亲和素─卵白素─生物素复合物 (SABC)法检测 40例大鼠皮肤辐射损伤模型标本EGFR的表达 ,实验动物按照射剂量分为 4个剂量组。结果 5Gy组、15Gy组、30Gy组、45Gy组EGFR阳性表达率分别为 2 7.0 %、49.3%、72 .2 %、87.6 % ,与正常对照组 (10 .8% )比较 ,均有显著差异 (均P <0 .0 1)。另外 ,各照射组间差异亦显著 (P <0 .0 1) ,表现为随着照射剂量的增加 ,EGFR阳性表达率亦增加。结论 辐射可显著增加皮肤组织中EGFR的表达 ,可能为辐射激活并扩增c -erbB - 1基因的结果 ;并在一定照射剂量范围内 ,随着照射剂量的增加 ,c -erbB - 1基因的激活与扩增增强 ,这可能成为皮肤溃疡难以愈合的重要因素之一。
Objective To investigate the effect of radiation on the expression of epidermal growth factor receptor (EGFR). Methods The immunohistochemical streptavidin-avidin-biotin complex (SABC) method was used to detect the expression of EGFR in the skin of 40 irradiated rats. The experimental animals were divided into 4 groups according to the dose of irradiation. Results The positive rates of EGFR in 5Gy, 15Gy, 30Gy and 45Gy groups were 2 7.0%, 49.3%, 72.2% and 87.6%, respectively, which were significantly different from those in normal control group (10.8%) All P <0. 01). In addition, there was also a significant difference between groups (P <0.01). The positive rate of EGFR expression increased with the increase of irradiation dose. Conclusion Radiation can significantly increase the expression of EGFR in skin tissues and may be the result of radiation activation and amplification of c - erbB - 1 gene.
