目的探讨五味子对去卵巢大鼠骨质疏松症的治疗作用及其机理。方法将59只雌性Wistar大鼠随机分为空白对照组、假手术组、模型组、阳性对照组及五味子组,其中五味子组11只,其余每组12只。摘除大鼠双侧卵巢1个月后,五味子组灌服五味子水煎剂(1g/ml),阳性对照组予己烯雌酚(0.008mg/ml)灌胃3个月(5.6ml/kg)。对不脱钙骨切片进行形态计量并检测胫骨成骨细胞(OB)和骨髓基质细胞(MSC)中护骨素(OPG)、核因子-κB受体活化因子配体(RANKL)蛋白表达。结果五味子组大鼠胫骨骨小梁体积百分比较模型组显著增高,除骨皮质类骨质平均宽度外,骨小梁吸收表面百分比、骨小梁形成表面百分比、骨小梁矿化率术较模型组均显著降低,OB及MSC中OPG蛋白表达明显增高(P<0.05或P<0.01),OB中RANKL蛋白表达明显降低(P<0.05)。结论五味子对卵巢切除大鼠骨质疏松症治疗作用机理之一是通过促进OB和MSC中OPG的分泌,使之与RANKL的结合增多,进而抑制破骨细胞的活性。
Objective To investigate the therapeutic effect and mechanism of Schisandra on osteoporosis in ovariectomized rats. Methods Fifty-nine female Wistar rats were randomly divided into blank control group, sham operation group, model group, positive control group and Wuweizi group. Among them, there were 11 in Wuweizi group and 12 in each group. One month after removal of bilateral ovaries from rats, Schisandra chinensis was fed with decoction of schisandra (1 g/ml) and positive control was given with diethylstilbestrol (0.008 mg/ml) for 3 months (5.6 ml/kg). Morphometric measurements were performed on non-decalcified bone slices and the expression of osteoprotegerin (OPG) and nuclear factor-kappa B receptor activating factor ligand (RANKL) in tibial osteoblasts (OB) and bone marrow stromal cells (MSC) were detected. Results The percentage of tibial trabecular bone volume in the Schisandra chinensis group was significantly higher than that in the model group. Except for the average width of cortical bone, the percentage of trabecular bone absorptive surface, percentage of trabecular bone formation surface, and trabecular mineralization rate were compared. All groups were significantly lower, OPG protein expression in OB and MSC was significantly higher (P<0.05 or P<0.01), and RANKL protein expression was significantly decreased in OB (P<0.05). Conclusion One of the therapeutic mechanisms of Schisandra on osteoporosis in ovariectomized rats is to promote the secretion of OPG in OB and MSC, increase its binding to RANKL, and then inhibit the activity of osteoclasts.
