目的:对六种枳壳药材进行遗传多样性和亲缘关系研究。方法:采用AFLP技术,随机选用10对EcoRI/MseI引物组合构建样品DNA指纹图谱。结果与结论:扩增产物共检测到清晰条带545条,多态性条带502条,特异性条带226条。有两对引物组合扩增条带多态率为100%,平均多态率为92.11%。利用引物组合E-ACA/M-CTA和E-ACG/M-CAA的特异性扩增带型可将各样品区别开来。此外,以相似系数0.5340为阈值,将六个枳壳品种聚类结果分为三类:黄土岗画红和鸡子橙遗传关系最近首先聚在一起,然后和新干臭橙聚为第一类,同属地的臭橙和勒橙聚为第二类,香橙单独聚为一类。
Objective: To study the genetic diversity and genetic relationship of six kinds of Fructus aurantii. Methods: AFLP technique was used to randomly select 10 pairs of EcoRI / MseI primer combinations to construct DNA fingerprinting. RESULTS AND CONCLUSION: A total of 545 clear bands, 502 polymorphic bands and 226 specific bands were detected. There were two pairs of primer combinations amplified polymorphic bands of 100%, the average polymorphism was 92.11%. Each sample can be distinguished using a specific banding pattern of primer combinations E-ACA / M-CTA and E-ACG / M-CAA. In addition, the clustering results of six Citrus aurantium cultivars were divided into three groups based on the similarity coefficient of 0.5340: the genetic relationship between Huangtuganghuhong and Jizi Orange was first clustered together, The same ground orange and orange together into the second category, oranges together into a single category.
