首次刺激剂在产生肿瘤坏死因子中的作用

人们知道某些细菌细胞及其产物具有抗肿瘤作用已经100多年了.非经肠道给予实验动物或病人细菌产物,诸如脂多糖(LPS),可导致肿瘤内的严重出血反应.然而给予有效剂量的LPS常可引起循环性虚脱,以至死亡.但以其它免疫刺激剂处理,则往往可以见到肿瘤的消退.1975年Carswell报道,在经内毒素处理的荷瘤小鼠血清中存在着一种使肿瘤坏死的因子,并称之为肿瘤坏死因子(tumor necrosis factor,TNF).它可引起体内肿瘤的出血性坏死,对体外培养的癌细胞也具有细胞毒作用.TNF被认为是由巨噬细胞产生的.可以从内毒素休克的预致敏(Presensitized)动物血清中提取.LPS则属于TNF的刺激剂.本文的目的在于研讨TNF产生的条件及其动力学.结果表明,在有脂多糖存在的条件下,某些活化剂(Prming agents)诸如BCG、短小棒状杆菌及酵母聚糖(Zymosan)等,可以刺激机体的网织内皮系统(RES),因而是极好的TNF产生的诱导物.但这些诱导物在刺激产生的TNF方面有较大的差异性,主要取决于对RES的刺激程度.某些可诱导RES增生的物质,如痤疮丙酸菌(Propionibacterium acnes)和厌氧杆菌(Corynebacteriumanaerobium)因可诱发明显的RES增生,因而也可诱发高水平的TNF活性.然而PPD、OK432、PSK以及Choreito并不能诱发TNF的活性.鉴于上述观察,作者认为凡能诱导高水平RES增生的活化剂,均可诱发高水平的TNF活性.由此推论,痤疮丙酸菌是最好的TNF诱导剂.
