对5-羟色胺(5-HT)抗独特型单克隆抗体的功能进行鉴定.方法:细胞培养法,免疫组织化学ABC法和形态计量法.结果:培养平滑肌细胞显示5-HT受体免疫反应阳性,在正常培养基培养的平滑肌细胞的平均长度为(18.8±35.4)μm.培养基中含不同浓度的5-HT抗独特型单克隆抗体(分别为0.01%,0.03%,0.10%,0.30%,1%抗体原液),平滑肌细胞的平均长度分别缩短为(175.6±43.3)μm,(16.2±46.2)μm,(116.9±25.8)μm,(89.5±22.8)μm,(79.9±21.6)μm,除含0.01%抗体原液组外均比正常组有非常显著的差别(P<0.01).结论:该抗独特型单克隆抗体具有与5-HT相类似的诱导平滑肌收缩的功能,5-HT受体可能介导了这种功能.
The function of serotonin (5-HT) anti-idiotypic monoclonal antibodies was identified. Methods: Cell culture method, immunohistochemical ABC method and morphometric method. Results: The cultured smooth muscle cells showed positive immunoreactivity of 5-HT receptor. The average length of smooth muscle cells cultured in normal medium was (18.8 ± 35.4) μm. The medium contained varying concentrations of 5-HT anti-idiotypic monoclonal antibodies (0.01%, 0.03%, 0.10%, 0.30%, 1% antibody stock, respectively) and the average length of smooth muscle cells (175.6 ± 43.3) μm, (16.2 ± 46.2) μm, (116.9 ± 25.8) μm, (89.5 ± 22.8) μm, 9 ± 21.6) μm. There was a significant difference (P <0.01) between the control group and the normal group except 0.01% antibody solution. CONCLUSION: The anti-idiotypic monoclonal antibody has similar functions to 5-HT in inducing smooth muscle contraction. 5-HT receptors may mediate this function.
