目的:研究不同剂量的镇肝熄风汤对自发性高血压大鼠(SHR)血压影响,以及对心血管MAPK/ERK通路中的丝裂原激活蛋白激酶(MAPK,Mitogen-activated protein kinase)蛋白和MAPK mRNA表达的影响。方法:24周龄自发性高血压大鼠(SHR),雄性,50只,模型组、中药低剂量组、中药中剂量组、中药高剂量组、复方罗布麻组,每组10只,同源雄性WKY大鼠10只作为对照组,灌胃给药5周后,测量大鼠尾部血压,Western Blot方法测定心血管MAPK p38蛋白表达,用Real-Time RT-PCR(Real-Time Quantitative RT-PCR)方法测定大鼠MAPK mRNA基因表达。结果:镇肝熄风汤中药中剂量、高剂量组和复方罗布麻组与模型组比较,血压明显下降(P<0.01);镇肝熄风汤中药中剂量、高剂量组与对照组比较,MAPK p38蛋白表达无明显变化(P>0.05);镇肝熄风汤高剂量组与对照组比较,MAPK mRNA表达无明显变化(P>0.05)。结论:镇肝熄风汤有明显降压的作用,镇肝熄风汤中剂量组、高剂量组中的MAPK 38p蛋白表达和MAPK mRNA表达明显升高,促进血管内皮细胞的活性。
Objective: To study the effects of different doses of Zhengan Xifeng Decoction on blood pressure in spontaneously hypertensive rats (SHRs) and the effect of Mitogen-activated protein kinase (MAPK) on cardiovascular MAPK / ERK pathway And MAPK mRNA expression. Methods: 24-week-old spontaneously hypertensive rats (SHR), 50 male, model group, low dose Chinese medicine group, medium dose group, high dose group, compound Apocynum group, Ten male WKY rats were used as control group. The rats’ tail blood pressure was measured 5 weeks after intragastric administration. The expression of MAPK p38 protein was detected by Western Blot. Real-time RT-PCR ) Method was used to determine the gene expression of MAPK mRNA in rats. Results: Compared with the model group, the blood pressure of Zhengan Xifeng decoction of traditional Chinese medicine at medium dose, high dose and compound Apocynum venetum obviously decreased (P <0.01). Compared with the control group, MAPK p38 protein expression had no significant change (P> 0.05). Compared with the control group, Zhengan Xifeng decoction had no significant change in the expression of MAPK mRNA (P> 0.05). Conclusion: Zhengan Xifeng decoction has a significant antihypertensive effect. The levels of MAPK 38p protein and MAPK mRNA in Zhengan Xifeng decoction medium and high dose group were significantly increased, which promoted the activity of vascular endothelial cells.
