采用高效液相离子交换法对电泳纯SIEA 28 kDa抗原组分进一步分离纯化。对获得的色谱纯SIEA 28 kDa抗原分子进行薄层等电聚焦分析,测定其等电点。结果证明电泳纯SIEA 28 kDa抗原分子经高效液相离子交换法纯化后,280nm、220nm波长色谱图均为一个两侧对称、光滑的高峰曲线,色谱洗脱液对其免疫活性光密度图也为一个两侧对称、光滑的高峰曲线。色谱纯化证明SIEA 28 kDa抗原组分可能为单一分子,其等电点范围在p15-p17之间。
The components of electrophoresis pure SIEA 28 kDa antigen were further isolated and purified by high performance liquid chromatography. The chromatographic pure SIEA 28 kDa antigen molecules were analyzed by thin layer isoelectric focusing, and its isoelectric point was measured. The results showed that electrophoresis pure SIEA 28 kDa antigen molecules purified by high performance liquid chromatography, 280nm, 220nm wavelength chromatograms are a bilateral symmetrical smooth peak curve, chromatographic eluate for immunocompetent optical density is also A symmetrical, smooth peak curve on both sides. Chromatographic purification proved that the SIEA 28 kDa antigen component may be a single molecule with an isoelectric point in the range of p15-p17.
