目的探讨五指毛桃水提液对60Coγ射线损伤小鼠组织DNA的防护作用。方法 40只SPF级雄性小鼠随机分为5组,阴性对照组给予生理盐水,单纯照射组作为辐射模型组,3个低、中、高给药+辐射组分别给予相当于生药5、10、20g·kg-1·d-1的五指毛桃水提液,连续灌胃到第7天。除阴性对照组外,所有小鼠接受全身一次性60Coγ照射,照射剂量为6Gy。照射后第2天处死小鼠,取肺、肝、睾丸、股骨和外周血,分别制作单细胞悬液,用单细胞凝胶电泳实验进行DNA损伤检测。结果辐射模型组小鼠组织细胞DNA的尾部DNA百分率(tail DNA%)和尾矩高于阴性对照组(P<0.01),各辐射+给药组的tailDNA%和尾矩均显著低于辐射模型组(P<0.01),且随着给药剂量的增加,tail DNA%和尾矩降低。结论五指毛桃水提液对60Coγ射线损伤小鼠组织DNA具有一定的防护作用。
Objective To investigate the protective effect of aqueous extract from Peach Blossom on the DNA of tissues damaged by 60Co γ -ray in mice. Methods Forty SPF male mice were randomly divided into five groups. The negative control group was given normal saline. The radiation group was used as the radiation model group. The three low, medium and high dosage + 20g · kg-1 · d-1 five finger peach water extract, continuous gavage to the first 7 days. Except for the negative control group, all mice received whole-body 60Coγ irradiation at a dose of 6 Gy. On the second day after irradiation, the mice were sacrificed and the lung, liver, testis, femur and peripheral blood were taken out to make single cell suspension. The DNA damage was detected by single cell gel electrophoresis. Results Tail DNA% and tail moment of DNA in the radiation group were higher than those in the negative control group (P <0.01), and the tail DNA% and tail moment of the radiation + treatment group were significantly lower than those in the radiation model Group (P <0.01), and tail DNA% and tail moment decreased with the increase of dose. Conclusion The five-finger peach water extract has some protective effects on the tissue DNA of 60Coγ-ray-injured mice.
