目的了解抗氧化剂阿魏酸钠对H2O2诱导人淋巴细胞凋亡过程中形态及生化的变化。方法采用电镜,激光共聚焦显微镜,MTT及DNA电泳分析方法。结*果H2O2处理细胞后Bcl-2基因表达明显减少,加入阿魏酸钠后明显恢复,而H2O2对Bax基因表达略有增加,加入阿魏酸钠后明显降低。结论H2O2能诱导淋巴细胞凋亡,阿魏酸钠可减轻细胞凋亡。同时阿魏酸钠可使H2O2诱导的细胞Bcl-2基因表达明显恢复,对Bax基因表达明显降低。
Objective To investigate the morphological and biochemical changes of antioxidant sodium ferulate in H2O2-induced human lymphocyte apoptosis. Methods Electron microscopy, laser confocal microscopy, MTT and DNA electrophoresis analysis methods. The results showed that the expression of Bcl-2 gene was significantly decreased after H2O2 treatment. The expression of Bcl-2 gene was significantly restored after addition of sodium ferulate, while H2O2 increased the expression of Bax gene slightly. Conclusion H2O2 can induce apoptosis of lymphocytes and sodium ferulate can reduce apoptosis. At the same time, sodium ferulate could obviously restore the expression of Bcl-2 gene induced by H2O2 and reduce the expression of Bax gene significantly.
