Ku蛋白是由Ku70和Ku80 2个亚基组成的异源二聚体结构,因对DNA末端的识别保护作用、对整个非同源末端修复复合体的成核作用而成为非同源末端修复途径有效性的主要决定因素,决定了细胞的损伤修复能力。基因组的稳态依赖于有效DNA修复机制,未被有效修复的DNA损伤是基因突变和肿瘤形成的重要机制。目前,有关Ku蛋白在肿瘤发生、发展中的作用存在多种观点。本文就Ku蛋白的特点及其在肿瘤DNA损伤修复中作用的研究进展作一综述。
The Ku protein is a heterodimer structure consisting of Ku70 and Ku80 2 subunits. It is a non-homologous end repairing pathway due to the recognition and protection of DNA ends and nucleation of the entire non-homologous end repair complexes. The main determinant of effectiveness determines the cell’s ability to repair damage. The homeostasis of the genome depends on effective DNA repair mechanisms, and DNA damage that has not been effectively repaired is an important mechanism for gene mutation and tumor formation. At present, there are many opinions about the role of Ku protein in tumorigenesis and development. In this paper, we reviewed the characteristics of Ku protein and its research progress in the role of DNA damage repair in tumors.
