目的探究miRNA-22对食管鳞状细胞癌(esophageal squamous cell carcinoma,ESCC)放射敏感性的影响。方法建立miRNA-22表达的稳定转染细胞模型,通过克隆形成实验和MTT法检测miRNA-22表达对受照食管癌细胞增殖活性的改变。结果体外稳定转染细胞模型中,克隆形成实验和MTT法结果表明,miRNA-22表达升高,显著降低了食管癌细胞增殖活性(P<0.01),促进了ESCC细胞对γ-射线的放射敏感性。反之,miRNA-22表达降低,显著增加了食管癌细胞增殖活性(P<0.01),抑制了ESCC细胞对γ-射线的放射敏感性。结论 miRNA-22的表达与食管癌放射敏感性密切相关,miRNA-22表达量越高,食管癌细胞放射敏感性越高。
Objective To investigate the effect of miRNA-22 on radiosensitivity of esophageal squamous cell carcinoma (ESCC). Methods A stable transfected cell model of miRNA-22 expression was established. The changes of miRNA-22 expression on the proliferation of esophageal carcinoma cells were detected by colony formation assay and MTT assay. Results Clone formation assay and MTT assay showed that the miRNA-22 expression was significantly increased and the proliferation of esophageal cancer cells was significantly decreased (P <0.01), and the radiosensitivity to γ-rays in ESCC cells was promoted Sex. On the contrary, miRNA-22 expression decreased significantly increased esophageal cancer cell proliferation (P <0.01), inhibiting ESCC cells γ-ray radiosensitivity. Conclusion The expression of miRNA-22 is closely related to radiosensitivity of esophageal cancer. The higher the expression of miRNA-22, the higher the radiosensitivity of esophageal cancer cells.
