分别采用摇床法和流通池法测定罗替戈汀微球的体外释放度,并通过与体内药动学曲线拟合进行选择。结果表明,摇床法和流通池法测定的体外释放度均与体内吸收具有明显相关性,相关系数r分别为0.9937和0.9978,且流通池法可检测到微球存在的较小突释。摇床法和流通池法均可用于罗替戈汀微球的体外释放度检查,流通池法作为一种新型溶出度检测方法,对指导处方筛选有较大意义。
The release of rotigotine microspheres in vitro was determined by shaker method and flow cell method, respectively, and selected by in vivo pharmacokinetic curve fitting. The results showed that both the shaker method and the flow cytometry in vitro release were significantly correlated with the absorption in vivo, the correlation coefficients r were 0.9937 and 0.9978, respectively, and the flow cell method could detect the small burst of microsphere present. Shaker method and flow cell method can be used for in vitro release of rotigotine microspheres examination, flow cell method as a new dissolution testing method, directed prescription screening has great significance.
